SFC protocol with heated flow cell and HC kept at room temperature, with  data collection, while the sample is held in the flow cell, was carried out in miniSIA-1 apparatus (A, B) identical to the one, used in previous page, but with the addition of the thermostated 12 cm long Garth cell.

With aim to maximize response, by optimizing temperature and incubation time in the flow cell, the temperature was varied while using  the SFC protocol shown on the next page. The difference between responses of sample (C 100ppB N) and blank (D) identified optimum conditions (45 C in FC and stop 40 sec, 21 C in HC), which were used when obtaining the calibration run (E).

The limit of sensitivity of this assay and  the reason for lag phase are discussed on next  page.
